Welcome Welcome to the ECSS 2010 Antalya Congress. ECSS 2010 Antalya Kongresine Hoş Geldiniz On behalf of the Middle East Technical University, Faculty of Education, Department of Physical Education and Sports and The European College of Sport Science (ECSS), we are privileged to welcome you in Antalya. We have prepared a stimulating scientific program in an excellent Mediterranean location for you. We hope you will enjoy both the science and the atmosphere of the 15th Annual Congress. The Theme of this year’s Congress is ‘Sport Science where the Cultures meet’. Sports play an essential role in uniting nations and cultures in the world with a unique inspiration. Having the 15th annual Congress of the ECSS in Turkey, a miraculous land where the cultures, religions and continents meet, emphasizes this years theme by also hosting many distinguished sports scientists from nations all around the world. With its constructive and pertinent content, the ECCS 2010 Antalya Congress shall be a valuable stage to inform and exchange ideas for the world sports science community. The scientific program of the congress is composed of more than 1300 abstracts, which were accepted after being carefully reviewed by the ECSS and the local scientific committees. The presented abstracts consist of all relevant sport science sub-disciplines and themes from an impressive diversity of 72 different nations. The abstracts are distributed among four Plenary Sessions, 36 Invited Symposia, 61 Oral Sessions and 6 Poster Sessions with more than 930 posters. Wednesday, June 23th, 2010 14:00 - 15:30 Oral presentations OP-BC01 Biochemistry 1 EXERCISE-ASSOCIATED CONCENTRATIONS OF IL-6 EXERTS A POSITIVE MODULATOR OF INSULIN SECRETION, SIGNAL TRANSDUCTION, NITRIC OXIDE RELEASE AND REDOX STATUS IN A CLONAL PANCREATIC B-CELL LINE. KRAUSE, M.1,2, MCCLENAGHAN, N.3, FLATT, P.3, MURPHY, C.2, NEWSHOLME, P.1 UCD-DUBLIN/ITT-DUBLIN Background and Aim: Interleukin-6 (IL-6) is a pleiotropic cytokine that influences metabolism in health and disease. IL-6 plasma levels are acutely elevated following muscle contraction and chronically during obesity. In this work we determined the effects of IL-6 (at various exercise-associated concentrations from 0 to 100 pg/mL) on pancreatic β-cell metabolism, AMPK signalling, insulin secretion, nitrite release and redox status in a rat clonal β-cell line. Methods: BRIN BD11 cells were incubated in the presence of IL-6 for 24h. Insulin secretion was determined by ELISA, signalling proteins by Western blotting and metabolites by spectrophotometric absorbance. Results: After 24hr incubation with IL-6, chronic insulin secretion (in μg/mg protein/24h) was increased from 128.78±7.3 (No IL-6) to 178.47±7.7 (at 100 pg/ml). Moreover, 24hr pre-treatment with IL-6 also altered both basal and nutrient-stimulated insulin secretion in subsequent 20min static incubations. Basal insulin secretion was increased 2-fold in the presence of 100pg/ml IL-6 (from 0.58±0.13 to 1.06±0.14μg/mg protein/20 min) whereas nutrient-stimulated insulin secretion increased 2-fold (from 1.5±0.09 to 2.9±0.3). Concomitantly, IL-6 enhanced both glutathione (GSH) and glutathione disulphide (GSSG) by nearly 20% without changing intracellular redox status (GSSG/GSH). IL-6 dramatically increased iNOS expression (by ca. 100-fold) with an accompanying 10-fold rise in nitrite release. AMPK levels were decreased by 43% whereas phosphorylated AMPK levels were elevated by 89% and CAMKK levels were increased by 50% after IL-6 exposure. Conclusions: We believe that IL-6 drives Ca2+ dependent signaling via CaMKK activation thus enhances FA oxidation in beta cells and decreases fatty acid synthesis, as CaMKK is an upstream kinase-activator of AMPK in the regulation of betacell metabolism and insulin secretion. In summary exercise-associated concentrations of IL-6 exerts positive effects on β-cell signalling, metabolism, anti-oxidant status and insulin secretion. The course of the insulin secretion during and after the exercise is, perhaps, related with IL-6 effects.

EXERCISE INDUCED ENDOCANNABINOID AND IMMUNE SYSTEM ALTERATIONS OZDURAK, R.H.1, KORKUSUZ, P.2, UCKAN, D.2, KILIC, K.2, KORKUSUZ, F.1 MIDDLE EAST TECHNICAL UNIVERSITY (1), HACETTEPE UNIVERSITY (2) Endocannabinoids play role in adaptive and innate immune systems through their receptors CB1 and CB2 located on immune effector cells. The aim of the study was to determine endocannabinoid and immune system alterations at moderate (18 m/min) and endurance (32 m/min) exercise intensities compared to controls. In the first part of the study, rats were exercised for 60 minutes/day, 5 days/week for 16 weeks. Immune effector cell proportions (T cells, B cells and NK cells) and endocannabinoid serum levels were determined by Flow cytometry and HPLC. Immunohistological analysis by TUNEL method and PCR was conducted in spleen and thymus tissue homogenates for apoptosis and cannabinoid system alterations. Anandamide (ANA) and 2 arachidonyl-glycerol (2-AG) serum levels increased with endurance type of exercise compared to moderate and control groups. mRNA expression of the CB1 receptor was also increased together with ANA in the same group. Apoptotic index increased in the endurance exercise group while immune effector cells responded divergently. CD45RA+ B cell percentage decreased while CD3+ T cell and CD161a+ NK cell percentage increased in blood. In spleen, however, CD8+ subtypes increased whereas CD11b+ cell and CD25+ cell numbers decreased in the spleen in the endurance type of exercise group. In the second part of the study, rats were grouped as the control, the endurance type of exercise, the DMSO, the AM281 (CB1 receptor antagonist) and the AM281+AM630 (CB1+CB2 receptor antagonist) groups. Immune effector cell proportions (CD3+ T cells, CD4+ T cells, CD8+ T cells, CD4+/8+ double positive T cells, activated T cells, CD45RA+ B cells and CD161a+ NK cells) in spleen and thymus were determined in addition to blood by flow cytometry. Endurance type of exercise associated significantly to immunological changes particularly to that of the T cells and their subtypes in blood and spleen. CD3+ T cells increased whereas CD8+ T cells decreased in blood. CD3+ T cell and CD4+/8+ double positive T cell percentages significantly increased in the spleen. Activated T cells and CD3+CD161a+ NK like T cells furthermore decreased in the spleen. AM281 and/or AM630 could partially reverse the effect of exercise in blood but not in the spleen. Alterations in the thymus were not observed. It can be concluded that endurance exercise causes immune system alterations through ANA and/or 2-AG. These alterations are CB1 and CB2 receptor dependent in blood, but not in splenic T cells and their subtypes. Endurance exercise triggered apoptosis in spleen which could also be reversed by CB1 and/or CB2 receptor antagonists.

Futsal players suffer injuries resulting from muscle fatigue and contact/collision among players. Muscle lesions can be detected by measuring muscle lesion markers such as CK and LDH [1]. After an initial lesion, the levels of acute-phase markers such as CRP and proinflammatory cytokines (TNF-a, IL-1b, IL-6 and IL-8) [2] increase in plasma. These inflammatory mediators may activate neutrophils and contribute to tissue damage, as well as increase susceptibility to invasive microorganisms [3], thus affecting the health of

OP-BC01 Biochemistry 1 players. In this study, we investigated the effect of a futsal match on muscle lesion markers, proinflammatory cytokines, and acute-phase protein of the players. We also evaluated the responsiveness of neutrophils after playing futsal based on the measurement of neutrophil death (necrosis and apoptosis), phagocytosis ratio and production of reactive oxygen species (ROS). Methods. Blood samples were taken from 16 futsal players (19±1 yrs, 68±6 kg, 51±2.6 ml•kg-1•min-1) before and immediately after a futsal match. Plasma cytokines and lesion markers were measured by ELISA and the enzymatic spectrophotometric method, respectively. CRP was determined by the immunoturbidimetric method. Neutrophil death (necrosis and apoptosis), phagocytosis ratio and ROS production were assessed using a FACSCalibur Flow Cytometer. Results. Exercise increased the serum levels of CK (2.5-fold), LDH (1.3-fold) and CRP (1.6-fold). Playing futsal also increased the serum levels of IL-6 (1.6 times). TNF-a, IL-1b, and IL-8 serum levels were not modified in the conditions studied. The futsal match induced neutrophil death possibly by apoptosis, as indicated by phosphatidylserine externalization. The exercise also increased the basal responsiveness of neutrophils, diminished the release of ROS by stimulated neutrophils, and decreased the phagocytosis capacity. Conclusion. Playing futsal induces inflammation and reduces the efficiency of neutrophils against infection resulting from exposure to pathogens. References. THE EFFECTS OF NORMOBARIC HYPOXIA ON MITOCHONDRIAL RESPIRATION IN RATS BISHOP, D., FERRI, A., RIVOLTA, I., PANARITI, A., ZAZA, A., MISEROCCHI, G. VICTORIA UNIVERSITY Introduction Contrasting mitochondrial changes have been reported in response to stays at high altitude. Some studies have reported increases in cytochrome c oxidase, while others have observed decreases in mitochondrial volume [1]. However, none of these studies directly measured mitochondrial respiration and all are limited by a lack of control for changes in physical activity following the ascent to high altitude living. The purpose of this study was to investigate the effects 30 days of normobaric hypoxia on ADP-stimulated mitochondrial respiration. Methods Twenty Wistar rats were randomly assigned to 30 days of either normobaric normoxia (CON; 21% O2) or hypoxia (HYP; 10% O2). Both submaximal (0.1 mM ADP) and maximal (2 mM ADP) ADP-stimulated mitochondrial respiration were determined on isolated mitochondria (from the lungs) and permeabilised muscle fibres from the left (LV) and right ventricle (RV), and the soleus (SOL) and extensor digitorum longus (EDL).
